Could anyone please tell me which kick start spring should be fitted to my 1957 16ms with amc box should it be an 'M' or 'MA' type.

Welcome, Peter.
The Bruce Main-Smith copy of the original AMC parts list is ESSENTIAL. Get a copy (0116 277 7669 if that's still their number). It has exploded diagrams of your machine and the AMC part numbers of all components; numbers which all spares suppliers use.
You will fumble around in the dark forever more without it. Believe me.

just checked my spares list and the partno for the spring was the same, 040044, from 1958 to 1963 for the AMC gearbox. So the M/MA designation must indicate something else, perhaps different gear ratios or different applications. My 1963 G12 has the MA pre-fixed gearboxnumber.

If your gearbox is pre 62 you should have the “M” type, this will be stamped on the top of the case before the number.

There are examples in the “Photo” section that should help you identify the part, (if you remove the inspection cover the spring will be visible):
